Controlled rate freezers can be used to prepare samples prior to cryogenic storage, ensuring maximum viability. These units are great for a number of reasons, notably that they are low energy and emissions operators: because LN2 storage depends on the cooling power of naturally sourced nitrogen gas, there are no halogenated hydrocarbon refrigerants used and no greenhouse gas concerns.

Thermo Scientific CryoMed freezer from American Instrument Exchange.

The Thermo CryoMed model 7452 is a front loading unit with a 34L capacity and a chamber temperature range from 50°C to -180°C. It features an integrated control system (consisting of freezing chamber, microprocessor control and printer – all in one bench top unit), allowing multiple freezing profiles to be tailored to specific protocols. Built-in dual solenoid valves increase LN2 injection volume for precise temperature control and accelerated freezing. This unit is specifically designed air handling system and patented liquid nitrogen injection ring work in tandem for precise temperature control and uniformity while over-temperature protection for heating provides a safe working environment. Standard alarms provide for open thermocouple, heater malfunction, high/low temperature limit, temperature tracking, power failure and completion of run.

Six pre-set profiles are factory programmed while an additional ten user defined profiles can be programmed by the customer. These profiles included:

  • Pre-Set Profile #1 – Commonly used for 2.0ml sample size, resulting in a 1°C rate from nucleation to -40°C and a 10°C per minute cooling rate to a -90°C end temperature.
  • Pre-Set Profile #2 – Commonly used for very small (96 well) sample size, resulting in a 1°C rate from nucleation to -40°C and a 10°C per minute cooling rate to a -90°C end temperature.
  • Pre-Set Profile #3 – Commonly used for straw size samples, resulting in a slow rate to -7°C, a 5 minute hold for manual cell nucleation, a 0.3°C rate to -35°C, and then a 0.1°C per minute cooling rate to a -37°C end temperature.
  • Pre-Set Profile #4 – Commonly used for hemopoietic stem cells that are 65-100ml in size, resulting in a 1°C rate from nucleation to -45°C and a 10°C per minute cooling rate to a -90°C end temperature.
  • Pre-Set Profile #5 – Commonly used for skin packets, resulting in a 1°C rate from nucleation to -35°C and a 10°C per minute cooling rate to a -90°C end temperature.
  • Pre-Set Profile #6 – Commonly used for hemopoietic stem cells that are 65-100ml in size, resulting in a 0.5°C rate from nucleation to -20°C and a 1°C per minute cooling rate to a -80°C end temperature.

We pretty psyched about this Hypoxia Chamber from Plas Labs. The model 856-HYPO is ideal for tissue culture work, including tumor cell and stem cell research, and is engineered to provide a controlled temperature, oxygen and carbon dioxide work area. It features an internal circulation fan, single electrical receptacle, and clear acrylic transfer chamber. One pair of glove ports are mounted on the main chamber.

The unit is controlled via the 8” color touch screen display mounted on the fan and motor housing, where it is easy to see from the operator’s position. The main screen displays the chamber process values for oxygen and carbon dioxide percentages and temperature. Oxygen can be controlled from 0-25%, carbon dioxide from 0-20%, and temperature from ambient to 40°C. The display also features on-screen data logging and external data logging (to hard drive or USB thumb drive). Other features of the control system include a 240-watt silicone heater, internal circulation fan, serrated gas inlets for nitrogen and carbon dioxide, an internal linear pump for oxygen introduction, and internal metering valves for nitrogen and carbon dioxide.

Hypoxia Chamber.

The control system is connected to sensors on the back of the Hypoxia Chamber that constantly send an analog signal back to the internal programmable logic controller (PLC). The PLC converts the respective analog signals into oxygen and carbon dioxide percentages. Depending on the set points, the PLC will either remove or add oxygen or carbon dioxide.

The basic housing of the Plas Labs 856-HYPO is formed of .375” thick clear cast acrylic and bright white acrylic back and floor with one piece thermoset white bottom, one pair of glove port rings with gloves, LED light strips (top mounted), pressure relief valve, internal circulation fan and single electrical receptacle with sealed cover.

The transfer chamber is constructed of .375” thick acrylic plastic which is also rigid. The clear acrylic provides constant visual observation over any items being transferred into or out of the glove box and is mounted to the right side of the Hypoxia Chamber. It includes inner and outer doors with black neoprene gasket, continuous stainless-steel hinge and two stainless steel toggle clamps; two gas purging valves, and vacuum gauge.

If you’re looking for a cost effective glove box for tissue culture work, please give us a call or send an email today about the Plas Labs model 856-HYPO Hypoxia Chamber. Thanks!
